چکیده
Abstract Study question In Individuals with raised Sperm DNA Fragmentation Index (DFI), will sperm selection by magnetic activated cell sorting (MACS) or Microfluidic Sorting (MF) optimize the reproductive outcomes? Summary answer MACS MF offer comparable outcomes. There seems no superiority between them. spermDFI, MACS/MF can be considered to results. What is known already It evident that DFI negatively affects Management for outcomes still elusive. design, size, duration This an ongoing Randomized Control Trial (RCT) prior approval from institutional Ethical Committee (IEC) (ECR/1312/Inst/TG/2019). preliminary data of this study. 6months(July 2020–January 2021). Couples undergoing IVF stimulation spermDFI were randomized into MACS(n = 75) and MF(n 58) selection. study also intends look at efficiency reduce post intervention. trial registered Clinical Trials Registry India (CTRI) REF/2021/05/043439 Participants/materials, setting, methods one failed offered DFI. DFI>30% recruited. testing was done SCSA method using software. processing based on randomization day Oocyte Retrieval(OPU). as per manufacturer’s instructions. Culture till blastocyst stage freeze all policy followed. Two Blastocysts showed100% survival transferred in a Frozen Embryo transfer (FET) cycle. Main results role chance Implantation rates(IR) Miscarriage Rates (MR), Multiple Pregnancy Rates(MPR) Live Birth rates (LBR) compared groups Reduction value evaluated arm - 75 subjects recruited, 49 patients (65%) had atleast 2 blastocysts frozen underwent FET. 26 (35%) cancelled cycles due formation. 58 recruited 38 20 reduction Vs 83% men reduced < 30% intervention 87% Both seem efficient separate sperms good quality Reproductive Outcomes IR 50% 48% (p 0.8540) MR 5.3% 11% 0.3280) MPR 8% 18% 0.1624) LBR ET cycle 63% 1.0000) comparable. Data doesn’t prove over another Limitations, reasons caution presented here small sample size Wider implications findings Optimal management needs further research. However, both are non invasive Nevertheless while offering these aspects like cost, time taken process technical challenges if any, should appropriate counselling. registration number

P-240: Effect of Oxidative Stress on DNA Fragmentation Index in Sperm of Mature Mice
Background: One of the major factors affecting on male infertility is oxidative stress (OS). Two main sources of ROS production are leukocytes and spermatozoa. The ROS such as tertiary-butyl hydroperoxide (TBHP) may damage and fragmentate to sperm DNA, which can lead to complications in the offspring. In this study, we will investigate the effect of od on DNA fragmentation index (DFI) in sperm ...

The editors of this book, Drs. Ashok Agarwal, Ahmad Majzoub, Chak-Lam Cho and Sandro Esteves, are to be congratulated for bringing together an extensive tome on the evaluation and management of sperm DNA fragmentation. The perspectives from North America, South America, Europe, Asia and the rest of the world provide an interesting snapshot into management of the unique phenomenon of sperm DNA d...

(1) that sperm DNA fragmentation (SDF) should be included in the evaluation of male factor infertility, along with semen analysis. Although there exist divergent views on this topic, our group has routinely employed SDF (TUNEL assay) and other methods for identification of sperm DNA damage ...
